DNA Replication is the process by which a DNA molecule produces two identical DNA molecules before cell division.
DNA replication begins at a specific site called the origin of replication. The enzyme DNA Helicase breaks the hydrogen bonds between the two strands and uncoils the DNA, forming a replication fork. Since DNA molecules are very long, replication occurs only in small regions at a time, not along the entire length.
Each parental DNA strand acts as a template for the formation of a new strand. The enzyme DNA Polymerase adds nucleotides complementary to the template strand according to base-pairing rules (A-T and G-C).
Replication is fast and highly accurate because DNA polymerase checks and corrects mistakes during synthesis. The process is energetically expensive as it requires energy supplied by nucleotide triphosphates. It is also substrate-dependent, requiring deoxyribonucleotides, and enzyme-dependent, involving several enzymes such as helicase, polymerase, and ligase.
Finally, two identical DNA molecules are produced, each containing one parental strand and one newly synthesized strand. This type of replication is called Semiconservative Replication.
